This project focuses on developing synthetic strategies for creating well-defined ubiquitin conjugates, which are crucial for biological research and therapeutic applications. It aims to overcome limitations of existing enzymatic methods by exploring chemical approaches.
Ubiquitination and deubiquitination of proteins has emerged as an important biological regulatory process.
For any investigational and therapeutical purposes, well-defined ubiquitin(-like) conjugates are of prime importance and to date, these have mainly been constructed by enzyme based strategies.
However, besides the general potential problems of enzymatic transformations (e.g. yield, isolation, expression), the promiscuous nature of ubiquitination severely limits their general applicability.…
