This project explores the mechanisms of DNA double-strand break repair, particularly in B lymphocytes. It focuses on how DNA end processing influences repair outcomes, aiming to understand the balance between protective and resection-promoting factors to better comprehend immunodeficiencies and cancer predisposition.
This proposal addresses a fundamental issue in molecular biology: how is repair of DNA double-strand breaks (DSBs) steered towards the appropriate physiological outcome? DSBs are cytotoxic DNA lesions that arise as a by-product of DNA replication, but also as a physiological intermediate during antigen receptor diversification in the Immune system. DNA end processing is a major determinant of DSB repair outcome.
